The Induction of Cellular Senescence as a Novel Anticancer TherapyFunding:Principal Investigator:Lab Website:(Lab website not available at this time) Project Summary:With the demonstration that prostate cancers are chemosensitive, efforts are being directed toward targeting these therapies in a rational manner. Recently, it has been recognized that a mechanism of terminal proliferation arrest after chemotherapy involves the reactivation of senescence. We have recently characterized a number of novel pathways, including that lead to senescence in human prostate epithelial cells. In this proposal, we will apply these results to define the reactivation of senescence pathways using clinical therapies for prostate cancer in human xenografts. We will determine if the senescence phenotype can be augmented to enhance the therapeutic efficacy of currently available chemotherapeutics. If successful, this information will be rapidly translated into clinical human trials that utilize senescence both as a marker of response and for directing therapy.
